High-throughput platform for High Content Screening image acquisition and high-content analysis. It enables automated acquisition, analysis, and management of fluorescence, brightfield, and digital phase contrast images in multiwell plates, providing a powerful solution for cellular phenotyping and quantitative studies. It supports the study of both live and fixed cells, the characterization and analysis of subcellular compartments, as well as 3D spheroid imaging.

Description

Fully automated High Content Imaging system designed for the acquisition, analysis, and management of cellular experiments in in vivo and in vitro models, focusing on reproducibility, high resolution, and experimental scalability.

Optical and Acquisition System:

  • High-precision laser-based autofocus system, optimized for stability during long-term acquisitions.

  • Motorized high-resolution stage for scanning multiwell plates and slides.

  • Spinning disk confocal module with a pinhole system, designed for efficient out-of-focus signal reduction and contrast improvement in thick or dynamic samples.

High-power excitation LEDs:

  • UV: 365 nm | VIOLET: 440 nm | BLUE: 475 nm | CYAN: 510 nm

  • GREEN/YELLOW: 550 nm and 580 nm | RED: 630 nm | FAR RED: 660 nm

Emission filters: 430-500 nm, 500-550 nm, 570-650 nm, and 655-760 nm.

Objectives: Air objectives (5x, 10x, 20x) and water objectives (20x, 40x).

Compatible Sample Formats: Multiwell plates for bioimaging (standard SBS format) and slides (up to 4 simultaneously).

Detection System & Integrated Environmental Chamber:

  • 16-bit sCMOS camera (Resolution: 4.7 MP, 2160 × 2160 px; Pixel size: 6.5 µm). Fast acquisition mode up to 100 fps (948 × 948 px).

  • Integrated environmental chamber for maintaining physiological conditions during live-cell experiments: Controlled temperature (37–42 °C) and $CO_2$ control (1–10%).

  • Compatibility with fluorescence, brightfield, and phase contrast.

Robotic Sample Handling System:

  • Automated plate transfer between the microscope and stackers or incubator.

  • Stackers for SBS format plates with a capacity for 60 plates (fixed cells).

  • Automated incubator with a capacity for 44 plates (live cells).

Image Analysis Software (Harmony): Instrument control, offline analysis on an office computer, automated cell segmentation and analysis, advanced morphological analysis (STAR, textures), Machine Learning classification (PhenoLOGIC), 3D visualization and analysis, and intelligent rescanning (PreciScan).
