Equipment for the realisation of digital quantitative PCR tests.
The digital PCR technique is based on distributing the DNA sample in many 20.000-well individual PCR reactions within this equipment. DNA is diluted to a limit concetrantion, the reason why a molecule will be distributed in some’s partitions and won’t in others’. After the PCR reaction, the fluorescence generated in each partition is detected to identify positives and negatives. This distribution is adapted to a Piosson model with a correction for wells that received more than one DNA molecule, which allows to obtain an absolute value of the DNA concentration within the sample. Partitions in this equipment are made in a chip and an analysis of 24 chips can be carried out in the course of one experiment. It’s possible to use the SYBRGreen fluorophore as well as TaqMan hydrolysis probes.
