The PyroMark Q48 (Qiagen) is an automatized pyrosequencing equipment of high performance designed to offer quantitative, precise and reproducible analysis of DNA in applications such as methylation studies, SNPs genotype, detection of mutations and genic edition validation.

One of the main advantages of PyroMark Q48 is their capacity to evaluate methylation with an unprecedented resolution, allowing the perceptual quantification of methylation in each CpG place individually. This is crucial in epigenetic research, where small changes in methylation can have essential implications in genic regulation and diseases such as cancer, neurologic disorders and autoimmune diseases.

Main technical characteristics

1. Pyrosequencing of Technology

  • Based on detection in real time of the liberation of pyrophosphate (PPi) during the synthesis of DNA
  •  “sequencing-by-synthesis” system that generates luminescent signals that are proportional to the incorporation of nucleotides.

2. Sample capacity

  • 48 samples per run (48 bowls in plaque format), with a simultaneous processing for high efficiency.
  • Required volume of the sample: 10–20 µL (depending on the protocol).

3. Precision and Sensitivity

  • Resolution of a single nucleotide, ideal for applications such as:
    • Quantification of metalation in individual CpG places (detection range: 5–100%).
    • SNPs identification and punctual mutations with a sensibility of as far as <5% of minority alleles.
  • Reproducibility: Variation coefficient (CV) <5% in metalation measuring.
