Activation maps during ventricular fibrillation are constructed from the analysis of two-second time windows. Each map is classified into three categories according to its complexity: low (type I), comprising a single broad wavefront with no conduction block or areas of slow conduction; intermediate (type II), consisting of two wavefronts or a wavefront with areas of conduction block or slow conduction; and high (type III), comprising three or more wavefronts associated with areas of slow conduction and conduction block. The degree of complexity is expressed as a quantitative variable using the formula: CI = (number of type I maps x 0.1 + number of type II maps x 1 + number of type III maps x 2) / total number of activation maps.

The conduction velocity of the electrical impulse during ventricular fibrillation is the ratio between the distance between two electrodes located in a direction perpendicular to the isochrones and the difference between their activation times (average of 5 determinations). Only maps in which both the input and output wavefronts are identified at the edges of the activation maps are used to determine the velocity. The wavelength of the activation process during VF is calculated as the product of velocity and refractoriness.

Freeze-drying is a process in which the product is frozen and subsequently introduced into a vacuum chamber to perform the separation of water by sublimation. In this way, water from the solid to the gaseous state is removed from the environment without going through the liquid state. To speed up the process, freeze-sublimation cycles are used to practically eliminate all the free water contained in the original product, while preserving the molecular structure of the lyophilised substance.
We use Welch's method to obtain the power spectrum of the signals recorded with each electrode. The periodogram is calculated for the first four seconds of each signal (4096 points, 1 kHz sampling rate), fragmenting the recordings into 8 segments with 50% overlap and using the Hamming window. The mean dominant frequency (FD) in the scanned area is determined by averaging the maximum peak power spectral density between 5 and 48 Hz at each electrode. The mean spectral concentration (EC) is also calculated by averaging the percentage of the total energy contained in the FrD range ± 0.5 Hz in each recording channel.

High performance liquid chromatography (HPLC) equipment is used to separate the components of a mixture based on different types of chemical interactions between the analysed substances and the chromatographic column. The compound passes through the chromatographic column via the stationary phase (usually a cylinder with small rounded particles with certain chemical characteristics on its surface) by pumping liquid (mobile phase) at high pressure through the column. The sample to be analysed is introduced in small quantities with an autosampler and its components are differentially slowed depending on chemical or physical interactions with the stationary phase as they advance through the column. These components are quantified by a fluorescence detector that detects only those substances that emit fluorescent light. It’s used for naturally fluorescent compounds or fluorescent derivatives.

Optical mapping techniques allow the recording of simultaneous voltage and calcium variations characterising the action potential as well as calcium transients with high spatial resolution. Dual voltage and calcium recording is used to characterise transmembrane potentials and calcium variations in the same viewing plane. A light source consisting of a triplet of LEDs and a camera with high-speed electron multiplier charge-coupled charge coupled device (EMCCD) imaging sensors is used. The lens system and the switching circuit allow the recording of voltage and calcium parameters at the same time with the same camera. We record transmembrane voltage using a voltage-dependent fluorescent compound (di-8-ANEPPS) while Ca2+ is recorded with fura-4F. By obtaining phase maps and singularities we can characterise activation patterns and the degree of organisation of complex arrhythmias.

Activation times at each electrode are determined by identifying the moments of maximum negative slope of the ventricular electrograms. If the electrograms show two or more deflections, then the steepest slope of the activation complex is assigned as the local activation time. The mean of the consecutive activation intervals during ventricular fibrillation and the 5th percentile of the histogram of intervals between successive activations is determined for each channel of the multielectrode. We use the 5th percentile (shortest VV intervals) as an approximation of the functional refractory period during ventricular fibrillation.
